Abstract: | The problem of potato production in Indonesia is the production of high quality virus free propagules. The virus free potato propagules are derived from microtuber through tissue culture methods. The experiment was conducted in two steps: the first steps was the production of shoot on semisolid medium and the second steps was the addition of liquid medium. The first and the second medium had the same treatment combination of nitrogen and sucrose. The experiment was arranged in Completely Randomized Design consisting two factors. The first factor was nitrogen concentration (30, 60, 90 and 120 mM) and the second factor was sucrose concentration (30, 45, 60, 75 and 90 g/l). The result showed there was an optimum concentration of sucrose and combination between nitrogen and sucrose on the number of shoots and percentage of tuber dry weight. The treatment of 60 mM nitrogen and 90 g/l sucrose, 90 mM nitrogen and 75 g/l sucrose produced the highest dry weight percentage, fresh weight and size (diameter) of qualified microtubers. |
