Abstract: The objectives of this study were to get precise media components which were cheaper and easily found to produce in vitro sweet potatoes and to produce plants that grow faster and healthy. This experiment was done at the Laboratory of Biology and Molecular PAU IPB using sweet potatoes Sukuh variety. The factorial completely random design was applied, with treatments of fertilizers. Two different fertilizers were used Hyponex 20-20-20 and Terra-Novalgro, with concentrations of 1 g l-1 and 2 g l-1 for Hyponex and 0, 1 cc l-1, 2 cc l-1, and 4 cc l-1 for Terra-Novalgro fertilizer, respectively. Those concentrations of fertilizers were combined to get eight treatments and they were replicated five * and compared to the control. The result showed that the treatment of Hyponex 20-20-20 2 g l-1 and Terra-Novalgro 4 cc l-1 can be used to substitute nutrients to improve in vitro propagation medium for sweet potato because it has a significant effect on the development of internodes and leaf. On the other hand, treatment with Hyponex 20-20-20 2 g l-1 and Terra-Novalgro 4 cc l-1 did not increase the number of roots. This experiment also showed that the commonly used Murashige and Skoog (MS) media could be replaced because this in vitro propagation media is cheaper and easier to prepare.
