Abstract: Leaf tissues of oil palm from tissue culture have been proven to undergo DNA hypomethylation, while its flower tissues undergo hypermethylation. Changes in the genomic DNA methylation may result in changes of gene expression and genome instability. The objectives of this research were to detect the changes in the genomic DNA sequences among oil palm regenerants from the same clone, and to obtain unique bands as indicators for normal and abnormal plants. Plant materials were taken from plants of the clone MK 152 with normal fruits, heavily abnormal fruits (AbB), and severely abnormal fruits (AS2) collected from BPPT, Ciampea Bogor. Detection of genetic changes was conducted with RAPD (Random Amplified polymorphic DNA) technique using 10 random primers. The result of this study showed that changes in the DNA sequence in abnormal plants were detected with five primers. Three primers, i.e. OPC-08, OPC-09 and SC10-19 were able to distinguish between normal and abnormal plants with unique bands from each primer. OPC- 08 primer was able to distinguish plants with normal and abnormal flowers by a single unique band of 800-1000 bp present in normal plant. The two other primers showed unique bands in normal plant; however they also showed other unique bands which varied between AbB and AS2 regenerants. Genetic changes occurred among regenerants originated from the same clone.
